Uji Banding Pengukuran Protein Total Serum antara Metoda Tetes Layang, Refraktometer dan Spektrofotometer

Examination of serum total protein is often needed to assess the presence :' hypoproteinemia or hyperproteinemia in various cases, and the method of Examination commonly used is photometric (Biuret) or automatisation. In View of not all regions can make this instrument available, the method of Examination using Refractometer device and Flying-drops method can become alternatives for measuring serum total protein in remote region or in many Tablic health centres and small hospitals.This reseach is aimed to obtain the difference in the findings from serum protein measurement between Flying-drops method and Biuret method, between Tefractometer and Biuret, and between Flying-drops and Refractometer.
The materials for examination were 50 serum samples obtained purposely ^om in-patients and then total protein measurement was performed with three methods for each serum sample. Materials for examination were a series of cupric sulphate of various densities strarting from 1.100 ; 159.0 grams of CUS045H20 was solved in 1000 ml of water for Flying-drops method, Hand Jiefractometer device for Refractometer method, and Biuret kit with normal control serum (Precinorm U from BM) with Automatic Analyzer Hitachi 902 for Biuret method. Data obtained were analyzed by SPSS-PC using Anova ::atistical test.
This reseach shows There were significant differences between Flying-drops Hth Biuret method (p=0.000), between Refractometer with Biuret method >p=0.000), and between Flying-drops with Refractometer (p=0.000).
This reseach shows (statistically), there were significant differences be-tween the three methods (p = 0.000). Furthermore, it is necessary to consider replacing Biuret method with F/yrng-drops method or Refractometer.
Pemeriksaan protein total serum sering diperlukan untuk menilai adanya hipoproteinemia atau hiperproteinemia dalam berbagai kasus, dan metoda pemeriksaan yang sering digunakan pada umumnya secara fotometrik (biuret) atau otomatisasi. Mengingat tidak semua daerah bisa mengadakan peralatan ini, metoda pemeriksaan dengan alat Refraktometer dan metoda Tetes layang dapat merupakan alternatif untuk pemeriksaan protein total serum di daerah-daerah atau di banyak Puskesmas dan Rumah sakit kecil.
Penelitian ini bertujuan untuk mengetahui perbedaan hasil pengukuran protein serum antara metoda Tetes layang dengan Biuret, Refraktometer dengan Biuret dan Tetes layang dengan Refraktometer.
Bahan penelitian adalah 50 serum pasien rawat inap diambil secara purposif dan dilakukan pemeriksaan protein total dengan tiga metoda untuk setiap serum. Bahan pemeriksaan satu seri larutan kupri sulfat dari berbagai berat jenis berasal dari b.j 1,100; 159,0 g krital CuS045H20 dilarutkan dalam 1000 ml air untuk metoda Tetes layang, alat Hand Refractometer untuk metoda Refraktometer dan kit Bi¬uret dengan serum kontrol normal (Precinorm U dari BM) dengan alat Automatic analyzer Hitachi 902 untuk metoda Biuret. Data yang diperoleh dianalis dengan SPSS-PC menggunakan uji statistik Anova.
Penelitian ini menunjukkan terdapat perbedaan yang bermakna antara metode Tetes latang dengan Biuret (p=0,000), antara Refraktometer dengan Biuret (0,000 dan antara Tetes layang dengan Refraktometer (p=0,000).
Penelitian ini menyimpulkan terdapat perbedaan bermakna secara uji statistik dari tiga metode tersebut p=0,000). Perlu pertimbangan lebih jauh untuk menggantikan pemeriksaan protein total metoda Biuret dengan metoda Tetes layang atau refraktometer.
