Efek Ekstrak Daun Sirih (Piper betle L.) terhadap Penurunan Kadar Serum Asam Urat dan Ureum pada Tikus Putih

Peningkatan asam urat menjadi faktor risiko terhadap penyakit cardiovaskuler tradisional. Daun sirih (Piper betle L.) mengandung senyawa betlephenol diduga dapat menurunkan kadar asam urat. Penelitian ini bertujuan untuk membuktikan ekstrak daun sirih dalam menurunkan kadar serum asam urat dan ureum tikus putih jantan. Penelitian menggunakan tikus putih jantan berumur 3 bulan, berat 170-214 gram, dibagi 6 kelompok yaitu kelompok I kontrol, kelompok II (induksi dan dosis kombinasi ekstrak daun sirih : propolis (1:1)), kelompok III (induksi dan ekstrak daun sirih dosis 10,37 mg/200 gr BB), kelompok IV (induksi dan ekstrak daun sirih dosis 41,48 mg/200 gr BB), kelompok V (induksi dan allopurinol dosis 1,8 mg/200 gr BB), dan kelompok VI kontrol negatif (induksi dan aquadest). Induksi diet kaya purin yang diberikan adalah jus hati ayam sebanyak 3 ml secara oral selama sepuluh hari. Pengukuran kadar asam urat dan ureum menggunakan Spektrofotometri Biosystem A 15. Signifikansi diterima pada nilai P < 0,05. Hasil yang diperoleh dianalisis dengan uji ANOVA one-way dilanjutkan uji LSD test menunjukkan perbedaan asam uratsignifikan antara kelompok II dan IV dengan kontrol negatif. Namun, tidak memberikan pengaruh secara signifikan terhadap penurunan kadar ureum hewan coba. Ekstrak daun sirih dosis 41,48 mg/200 grBB dan dosis ekstrak kombinasi dapat menurunkan kadar asam urat, sehingga berpotensi sebagai bahan hipourikemia.
Hyperuricaemia is associated with an increased risk factors of traditional cardiovasculer diseases. The Betel (Piper betle L.) contain betlephenol may can used to decrease uric acid values. Its The aim of this study was to examined effect of that extract to decreased serum uric acid and ureum level on rat. Three month male Sprague-Dawley rats 170 to 214 g body weight were used for the experiments. High diet purin induction gave 3 mL/day for 10 days.Group I (withouth induction), Group II (induction and gave combine extract with propolis (1:1)), Group III (induction and extract 10,37 mg/200 gr BB), Group IV (induction and 41,49 mg/200 gr BB), Group V (induction and allopurinol 1,9 mg/200 gr BB), Gropu VI kontrol negatif (induction and aquadest). Spektrofotometric Biosystem A 15 were used to assesed levels of uric acid and ureum levels. P<0.05 was considered to indicate a significant difference. As a result Groups II and IV of experiment indicated new insights into the antihyperuricemic activities. Variance was analyzed using a one-way analysis of variance (ANOVA) and a LSD correction with statistical software. However no significant difference with ureum level. The Betel extract of 41,49 mg/200 g-bw doses and its combine with which can possibly be developed into potential hypouricemic agents.
