Proliferasi Limfosit Mencit BALB/c setelah Pemberian Ubi Jalar Ungu (Ipomoea batatas L.) Diinduksi Ovalbumin

Kandungan flavonoid dalam ubi jalar ungu (Ipomoea batatas L.) berkhasiat sebagai agen antialergi. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol I. batatas L (EEIB). terhadap proliferasi limfosit T pada mencit BALB/ct6e456rd6 diinduksi ovalbumin. Jenis penelitian ini adalah eksperimental post-test only control group design dengan subyek 28 ekor mencit jantan BALB/c dibagi 7 kelompok, yaitu kelompok kontrol (K), kontrol negatif diinduksi ovalbumin (KN), 4 kelompok perlakuan EEIB dosis 0.21, 0.42, 0.84, 1.65 g/kg bb/hari dan kelompok perlakuan obat anti histamin 0.02mg/20g bb/hari (OAH). Kelompok KN dan kelompok perlakuan diinduksi ovalbumin pada hari ke-15, hari ke-22 dan hari ke-23 sampai hari ke-28. Pada hari ke-29 dilakukan pembedahan, pengambilan organ limpa, dikultur, dan suspensi selnya dan dilakukan uji proliferasi limfosit T dengan metode ELISA. Data proliferasi limfosit T dianalisis dengan uji One Way ANOVA dilanjutkan uji Tukey. Hasil rerata proliferasi limfosit T tertinggi didapatkan pada kelompok induksi OVA (P1) sebesar 0.926 ± 0.145 dan terendah pada kelompok ekstrak etanol Ipomoea batatas L. dosis 0.84 g/kg bb/hari sebesar 0.562 ± 0.074. Hasil uji One Way ANOVA menunjukkan ada beda nyata (p=0.000). Disimpulkan bahwa Ipomoea batatas L. dapat menurunkan proliferasi limfosit T mencit BALB/c dengan dosis efektif 0.84 g/kg bb/hari.Flavonoids in purple sweet potato (Ipomoea batatas L.) have favorable properties such as antiallergy. This study aimed to determine the effect of etanol extract of Ipomoea batatas L (EEIB) on proliferation of T lymphocytes in BALB/c mice induced ovalbumin. This is an experimental research with post-test only control group design used 28 BALB/c mice which is divided into 7 groups: control group (C), negative control with ovalbumin induction (KN), EEIB treatment group doses of 0:21, 0:42, 0.84, 1.65 g/kg bb/day and antihistamines drug treatment group dose of 0.02mg/20g bw/day (OAH). KN group and treatment group are induced with ovalbumin on day 15th, day 22nd and day 23rd until day 28th. On day 29th, mice are decapited and dissected, their spleen are taken, being cultured, their cells are suspended, and do the proliferation of T lymphocytes tested with ELISA. Limfosit T proliferation data are analysed with One Way ANOVA followed by Tukey’s test. The result of the highest proliferation of T lymphocytes is obtained in the ovalbumin induction group (P1) of 0926 ± 0145 and the lowest is in Ipomoea batatas L. ethanol extract group dose of 0.84 g/kg bw/day of 0562 ± 0074. One Way ANOVA test results showed a significant difference (p = 0.000). It is concluded that Ipomoea batatas L. can decrease the T lymphocyte proliferation in BALB/c mice with an effective dose of 0.84 g/kg bw/day.
